Sedang asyik mengejar Joh Carter, mereka terserempak dengan orang India maka berlaku lah peperangan . Carter melarikan diri dan berlindung di dalam gua. Di dalam gua tersebut beliau menemui seorang memegang medalyon dengan kuasa telepati . Apabila Carter menyentuh, dia mendapati dirinya di tempat di mana beliau boleh melompat tinggi yang luar biasa, antara lain. Beliau kemudiannya menemui makhluk yang dia tidak pernah dilihat sebelum ini. Kemudian dia bertemu dengan seorang wanita yang membantu dia mendapati bahawa dia berada di planet marikh.

Nick Fury (Samuel L Jackson), yang merupakan pengarah agensi yang dinamakan S.H.I.E.L.D., tiba ke satu pusat penyelidikan yang sedang dalam proses pengosongan bangunan. Tesseract, sejenis sumber tenaga tidak terhad telah diaktifkan di pusat penyelidikan tersebut dan telah membuka satu laluan angkasa lepas lalu membolehkan Loki (Tom Hiddleston) datang ke Bumi melaluinya. Loki ialah adik Thor di dalam filem Thor. Loki mencuri The Tesseract dan menggunakan kuasa luar biasanya untuk mengawal minda beberapa orang ahli S.H.I.E.L.D seperti Ejen Clint Barton (Jeremy Renner) dan ahli fizik iaitu Dr. Erik Selvig (Stellan Skarsgard) untuk membantu beliau melarikan diri.

Sebagai tindak balas terhadap serangan Loki tersebut, Fury segera mengaktifkan Inisiatif the Avengers (the Avengers Initiative). Natasha Romanoff / Black Widow (Scarlett Johansson) telah pergi ke India untuk mencari dan merekrut Dr. Bruce Banner / Hulk (Mark Ruffalo), sementara itu. Ejen Philip Coulson (Clark Gregg) menemui Tony Stark /Iron Man (Robert Downey Jr.) dan memintanya untuk mengkaji kajian Dr. Selvig’s mengenai The Tesseract. Fury sendiri pula telah bejumpa dangan Steve Rogers / Captain America (Chris Evans) untuk memintanya membantu merampas semula the Tesseract daripada Loki. Semasa di dalam buangan, Loki berjumpa dan seterusnya bersekongkol dengan sekumpulan makhluk asing yang gemar berperang iaituChitauri. Chitauri bersetuju membantu Loki menakluk Bumi dengan syarat Loki menyerahkan Tesseract kepada mereka sebagai balasan kelak.

Iron Man, Captain America, dan agen Romanoff telah pergi ke Germany untuk menahan Loki yang sedang sibuk mencuri iridium iaitu sejenis bahan yang diperlukan untuk menstabilkan Tesseract. Selepas beberapa pergelutan dengan wira-wira ini, Loki pun menyerah diri dan seterusnya dibawa ke pesawat S.H.I.E.L.D. Walau bagaimanapun, Thor (Chris Hemsworth), iaitu abang angkat Loki telah merampas Loki dari tangan S.H.I.E.L.D dengan tujuan untuk menguruskan kejahatan Loki dengan caranya sendiri. Iron Man terpaksa berhadapan berdepan dengan Thor untuk mendapatkan Loki semula dari tangan Thor. Selepas beberapa pergelutan di satu hutan, Captain America segera tiba untuk meleraikan pergaduhan dan seterusnya mereka bersetuju untuk membawa Loki ke dalam sebuah Helicarrier. Di dalam Helicarrier tersebut, Loki telah dikurung ke dalam sebuah sangkar yang direka khas untuk mengurung The Hulk.

Di dalam Helicarrier, para Avengers berbincang akan bagaimana menguruskan Loki. Pada masa yang sama, Rogers menyedari bahawa S.H.I.E.L.D. sedang merancang untuk menggunakan Tesseract dalam pembinaan senjata yang paling dahsyat dalam sejarah manusia. Fury mengakui bahawa senjata yang terhebat tersebut adalah satu keperluan bumi kerana satu kejadian yang telah berlaku di New Mexico setahun sebelumnya telah menyedarkan S.H.I.E.L.D bahawa bumi adalah sangat lemah jika berhadapan dengan serangan makhluk asing. Sementara para Avengers sedang berbincang, Hawkeye, seorang ejen S.H.I.E.L.D yang telah dipukau oleh Loki menyerang Helicarrier dan merosakkan enjinnya. Iron Man dan Captain America terpaksa bergegas untuk menghidupkan semula enjin pesawat gergasi itu. Pada masa itulah Dr. Banner menjelma sebagai The Hulk. Thor terpaksa berkerja keras untuk menghalang Hulk daripada terus mengamuk tidak tentu arah. Dalam adengan inilah penonton boleh melihat betapa beratnya tukul Thor yang langsung tidak dapat digerakkan walaupun oleh Hulk.
